Adding a 100 amp shunt to a neg 4/0 awg for battery for my solar system. Will this work?

If the shunt has large enough area on it for the 4/0's ends then yes it will work.

That said if your using 4/0 cables I would think a 100 amp shunt will be a bit small amps wise.

100a is not much. What do you want to achieve?
What I'm trying to do is go from a upgrade my system to 24 volt ⚡️ and I have 2× 12v 200 ah batteries and about to get two more so I change the wire to 4/0 ... so I won't have to change them again if I go to 48v later.

If you haven't bought your shunt yet you might want to look at the victron 500amp smart shunt. Its going to be far more handy if you make it to 48v and works great at 24v and 12v too. Excellent shunt.

I'm using them on my 12v stuff and 48v stuff now and I'm in the process of even installing one on my electric riding mower :)

100 amp shunts have higher resistance. In U.S. they are typically 1milli-ohm. (100mv/100A)

For 4/0 cable, the 100A shunt resistance is equal to about 20 feet of 4/0 cable.

50mV/500A shunt is 0.1 milliohms of resistance.
